Toxicity of calcium hydroxide to human umbilical cord stem cells

Calcium hydroxide is the standard gold material which is routinely used in dental conservation treatments. This material is a white powder and has strong alkaline or strong alkaline properties. In regenerative endodontic therapy, the material is one of the preferred ingredient protocols because it can promote mineralization and accelerate healing process.

The use of stem cells in the field of tooth conservation is still minimal and needs to be investigated. Human umbilical cord stem cells are an attractive stem cell candidate for tissue regeneration therapy in the field of tooth conservation, incredibly regenerative endodontics.

Moreover, experimental studies were carried out at concentrations of calcium hydroxide from 0 to 25 micrograms/ml. This study was conducted to determine the response and as a basis for subsequent studies for regenerative dental treatment. Several studies have reported the toxicity test of calcium hydroxide on several types of fibroblast cells that have been standardized to show good biocompatibility. The research team was interested in carrying out this study because human umbilical cord stem cells have several distinct advantages over other cells. In this study, MTT test was conducted to determine cell life and determine the toxicity of calcium hydroxide to human umbilical cord stem cells.

After the research, calcium hydroxide at a concentration of 0 to 25 micrograms/ml showed that the higher the concentration of calcium hydroxide, the lower the number of living stem cells, meaning that stem cells can survive under conditions of calcium hydroxide at the concentration range. The shape and distribution of stem cells did not change under these conditions, but a decrease in the number of living stem cells occurred in the concentration range of 3.125 and 6.25 microgram/ml.

This research is expected to provide information on the prospect of stem cell utilization for the development of regenerative therapy in the field of tooth conservation, especially regenerative endodontics. (*)
